OP ended up on Reddit when her son asked her to help him when he was having financial problems.
OP’s daughter lives with her even though she is 29 since the economy isn’t great.
OP’s daughter does pay her parent’s rent monthly and isn’t dependent on her parents.
OP and his husband decided to save the rent she gives them and have been putting that money aside for years.
When OP’s son found out about the money, they wanted to have a discussion with OP about needing help.

I don’t see any sort of favouritism going on here.
While it is rent money, it is the OP and her husband’s choice what they want to do with it.
To be fair, OP did offer to help him financially.
It is clear that they are being greedy and want the amount that was saved by his parents for his sister.
He will not stop here if the OP gives in to this instance.
